Senolytics and Fisetin

At the center of this trend is senolytics.
As we age, senescent cells gradually accumulate in the body.
Senolytics are a smart anti-aging strategy that leaves healthy normal cells untouched,

while precisely targeting and removing only the aging cells that make the skin look older.

 

Among ten flavonoids, Fisetin was found to be the most effective compound for removing senescent cells.

Yes. In our experiment, skin aging markers visibly decreased by nearly half.
When the root cause of aging is reduced, it makes sense that the skin’s aging clock may slow down with continued use.

 

 

Senolytic Efficacy of Fisenolyme / Ex-vivo
The senescent cell-reducing effect of Fisenolyme was confirmed through p21 fluorescence staining.
After Fisenolyme treatment, the ratio of p21-positive cells decreased from 46% to 37.5%, showing an approximately 43% reduction.
While most nuclei in the control group showed strong red fluorescence, indicating a high level of senescence,

 

the density of red nuclei visibly decreased after Fisenolyme treatment.


 

Only the zombie cells, precisely targeted
Fisenolyme helps protect healthy cells, while accurately identifying senescent cells.
In the experiment, the signal that removes senescent cells became 26 times stronger than before.

 

 

In fact, this clearing signal was confirmed in 9 out of 10 senescent cells, or 90%.

 

 

Senolytic Efficacy of Fisenolyme / Ex-vivo
We evaluated whether Caspase-8, a key enzyme that directly induces apoptosis in senescent cells, was activated.
Unlike the control group, which showed no apoptotic signal, the Fisenolyme-treated group showed strong green fluorescence throughout the cytoplasm, proving activation of the apoptotic pathway.
Caspase-8 fluorescence intensity increased approximately 26-fold compared to the control group, quantitatively demonstrating excellent apoptosis-inducing efficacy.

 

 

To further verify whether Fisenolyme selectively eliminated senescent cells while protecting normal cells, two markers were merged and analyzed in detail.
In the Fisenolyme group, green apoptotic signals, Caspase-8, were clearly observed surrounding the remaining red senescent cell nuclei, p21+.
Notably, apoptotic signals were activated in as many as 90% of senescent cells, p21+, showing an approximately 98% increase compared to the control group and confirming precise targeting of senescent cells.

And it does not stop there.
After zombie cells are cleared away, the empty space is filled with collagen, the key to skin elasticity.

Anti-aging / Wrinkle-improving Efficacy of Fisenolyme
When treated with Fisenolyme at 10 μg/mL, the expression of COL1A1 and COL3A1,

 

 

the main collagen fibers in the dermis, increased by up to 85.2% and 98.0%, respectively.
In human dermal fibroblasts, it inhibited the expression of MMP1,

 

a wrinkle-inducing factor, by up to 50.5%, demonstrating excellent efficacy in suppressing skin aging.

 

 

 

Barrier-strengthening and Photoprotective Efficacy of Fisenolyme

Fisenolyme improves the expression of CerS3 and CerS4, genes essential for ceramide synthesis in the skin barrier,

 

suggesting potential support for long-term barrier function recovery.
Under UVA and UVB exposure, it reduces ROS generation and suppresses the expression of MMP,

 

 

MMP-1, and MMP-3, helping protect UV-damaged skin and prevent photoaging.
